Basic Principles and Purity Determination by Differential Scanning Calorimetry
Purity and quality are the major events in pharmaceutical sector. One of the most important tests that can be performed for pharmaceutical and organic material is the assessment of its purity by differential scanning calorimetry (DSC) during in process and finished product drug development. DSC has proven to be a rapid, accurate, precise and easy-to-use approach for the assessment of the purity of many different types of materials. This method depends only on the physicochemical behavior of the compound. This method is accurate for samples over 98% pure, but it does not measure impurities which are soluble in the solid phase or insoluble in the melt.
Differential Scanning calorimetry
Differential Scanning Calorimetry (DSC) monitors heat effects associated with phase transitions and chemical reactions as a function of temperature. In a DSC the difference in heat flow to the sample and a reference at the same temperature, is recorded as a function of temperature. The reference is an inert material such as alumina, or just an empty aluminum pan. The temperature of both the sample and reference are increased at a constant rate. The heat flow difference can be either positive or negative.
?dH/dT= [dH/dT]sample - [dH/dT]reference
Here dH/dT is the heat flow measured in mcal sec-1.In an endothermic process, such as most phase transitions, heat is absorbed and therefore heat flow to the sample is higher than that of the reference. Hence ?dH/dT is positive.
Features and benefits
For purity analysis, the power compensated DSC, provides a number of important features and benefits for the assessment of the purity of pharmaceutical and organic materials:
- Very low mass furnaces (1g) for sample side and reference side providing rapid response times critical for the accurate measurement of purities
- Measurement of true heat flow rather than temperature differential for more accurate calorimetric determinations
- Use PRT (platinum resistance thermometer) temperature sensors rather than thermocouples for the most accurate and precise measurement of sample temperature
- Outstanding resolution providing more theoretically correct melting responses (i.e., narrow peak half-width and greater peak intensity)
- Very high sensitivity
- Glass transition and melting point
- Crystallisation time and temperature
- Heats of melting and crystallization
- Percent crystallinities
- Oxidative stabilities and heat capacities
- Heats of cure and percent cure
- Polymorphism and recyclates and regrinds
- Compositional analysis
- Thermal stabilities and purities
The particle size of the sample also affected its purity value. The effects could be explained in terms of thermal lags, decomposition and evaporation during melting or sublimation before melting. Decrease of particle size may either lead to low purity values, due to concurrent increase in structural disorder or to increase in purity values, due to the formation of aggregates by electrostatic forces. It was concluded that if triturating of sample before subjecting to DSC analysis causes aggregate formation, purity values which are too high may result.
The purity of an organic substance can be estimated by DSC based on the shape and the temperature of the DSC melting endotherm. This procedure utilizes the Van’t Hoff equation, which is given as:
1/Fs = [?H/R]*[T0-Ts] T02*[1/X2]
Where Ts is the instantaneous sample temperature and the melting temperature K) and T0 is the melting temperature of the pure substance, ?H is the heat of melting of the pure substance (J/g), X2 is the mole fraction of impurity in the sample, R is a constant (8.314 J/mole) and Fs is the fraction of sample melting at temperature Ts. The fraction Fs is given as As/At where As is the area of the melting endotherm up to temperature Ts and At is the total area of melting endotherm.
